Enhanced Binding Analysis is a follow-on service to our Membrane Proteome Array (MPA) screening service. If your MPA screen returns an off-target binding event, this service provides deeper statistical, biological, and molecular insights for those off targets.  

These data comply with FDA guidance that “nonclinical evaluation of the antigen recognition domain should include assessment of the specificity and affinity/avidity for the target antigen/cells to evaluate the potential for on-target/off-tumor and off-target toxicities.” (FDA, 2024, Considerations for the Development of Chimeric Antigen Receptor (CAR) T Cell Products) 

An overview of Off-Target Risk Analysis.

With this service package, we can help you investigate:

  • The significance of off-target binding relative to your target; differentiating weak binding interactions and low off-target expression
  • The relative binding strength (affinity/avidity) of an off-target protein vs. your target protein, including EC50 and maximum binding (Bmax) calculations
  • Biological insights into sub-cellular and human tissue distribution of the off-target proteinto determine if the off-target is accessible to your drug

  • Test antibody run in triplicatefor the highest confidence, even with weak off-targets
  • 8-point, 2-fold titration with 4 replicates at each concentrationfor statistical analysis
  • Additional transfection, binding, isotype, and expression controls for comparison
  • Immunofluorescent imaging, permeabilization controls, and bioinformatic analysis for sub-cellular and tissue localization to determine if the off-target is accessible to your drug
